You have to unlock some of them with gold coins, the Minion and Angel are unlocked only for Premium Upgrades, two others are unlocked through achievements (endless mode and 2v2 PVP). Villains are playable characters that act similar to hero skins (but not the same). If you really want to help us out, buy the Premium Upgrade :) Otherwise, most people think Gax or Nameless are solid choices with Nameless being a little more advanced. For starting out, Gladius was designed to be a good all-rounder.
There's no correct answer for this because they are supposed to be balanced equally and have their own roles.
